Nuclear V-Type ATPase: Focus on “Vacuolar H-ATPase in the nuclear membranes

چکیده
In the accompanying article by Santos et al published in this issue of the American 14 Journal of Physiology: Cell Physiology (8) the authors tested the hypothesis that the V15 type ATPase was present and functional in the NE. The authors established that the 16 nuclear pH is acidic, pH gradients across the nuclear membrane were bafilomysin and 17 concanamycin sensitive and that the V-type ATPase was present in the NE by 18 immunolocalization, and in both the ONM and INM by western blots and ATPase 19 assays. 20

Vacuolar H+-ATPase in the nuclear membranes regulates nucleo-cytosolic proton gradients.
The regulation of the luminal pH of each organelle is crucial for its function and must be controlled tightly. Nevertheless, it has been assumed that the nuclear pH is regulated by the cytoplasmic proton transporters via the diffusion of H+ across the nuclear pores because of their large diameter. However, it has been demonstrated that ion gradients exist between cytosol and nucleus, suggesting...
متن کاملNuclear V-type ATPase. Focus on “Vacuolar H -ATPase in the nuclear membranes regulates nucleo-cytosolic proton gradients”
THE NUCLEAR ENVELOPE (NE) consists of two continuous membranes, with pores where the inner nuclear membrane (INM) meets the outer nuclear membrane (ONM) (7). V-type ATPases in the plasma membrane and intracellular membranes alter pH ( pH) and generate a transmembrane electrical potential ( ) across plasma membranes and intracellular compartments (6). In the accompanying article by Santos et al....
متن کاملMicroautophagy of the Nucleus Coincides with a Vacuolar Diffusion Barrier at Nuclear–Vacuolar Junctions
Nuclei bind yeast vacuoles via nucleus-vacuole (NV) junctions. Under nutrient restriction, NV junctions invaginate and release vesicles filled with nuclear material into vacuoles, resulting in piecemeal microautophagy of the nucleus (PMN). We show that the electrochemical gradient across the vacuolar membrane promotes invagination of NV junctions. Existing invaginations persist independently of...
متن کاملRegulation of vacuolar H+-ATPase activity by the Cdc42 effector Ste20 in Saccharomyces cerevisiae.
In the budding yeast Saccharomyces cerevisiae, the Cdc42 effector Ste20 plays a crucial role in the regulation of filamentous growth, a response to nutrient limitation. Using the split-ubiquitin technique, we found that Ste20 forms a complex with Vma13, an important regulatory subunit of vacuolar H(+)-ATPase (V-ATPase). This protein-protein interaction was confirmed by a pulldown assay and coim...
متن کاملRegulation and reversibility of vacuolar H(+)-ATPase.
Arabidopsis thaliana vacuolar H(+)-translocating pyrophosphatase (V-PPase) was expressed functionally in yeast vacuoles with endogenous vacuolar H(+)-ATPase (V-ATPase), and the regulation and reversibility of V-ATPase were studied using these vacuoles. Analysis of electrochemical proton gradient (DeltamuH) formation with ATP and pyrophosphate indicated that the proton transport by V-ATPase or V...
